I would say that depends on if the colleague is someone you actually support or not. If it is someone from outside the people you support, I would just say straightforward, I am so sorry, I am not going to have time to do that due to the other things I am working on. usually I suggest an alternative if I can (perhaps so and so would be able to assist you?)
If it is someone I support, I will say, I am working on xyz which is due at such and such a time. Can you check with so and so that gave me xyz to see if it can be bumped for your project? (I always try to let them fight it out instead of getting in the middle).
On the other hand, if it is my boss, I will just advise him, I can do that, but it will impact delivery of xyz (and then I notify so and so).
